  • Symposium | Advanced systemic mastocytosis: Diagnosis, risk stratification, and response assessment in 2026

    16:34|
At the European Hematology Association (EHA) 2026 Congress, June 11–14, 2026, Stockholm, SE, the MPN Hub held a symposium, titled Advanced systemic mastocytosis: Redefining the disease, rethinking patient outcomes. During the symposium, Johannes Lübke, University Hospital Mannheim, Mannheim, DE, delivered a presentation on diagnosis, risk stratification, and response assessment in advanced systemic mastocytosis (AdvSM).  Lübke discusses the current diagnostic framework for systemic mastocytosis, emphasizing the need for integrated disease assessment beyond established diagnostic criteria. He explores the role of molecular profiling in prognostic risk stratification, evolving prognostic models in the era of KIT-targeted therapy, current and emerging approaches to response assessment, and the complementary role of patient-reported outcomes in evaluating disease burden and treatment benefit. This educational resource is independently supported by Blueprint medicines. • Targeting KIT in advanced systemic mastocytosis

    26:01|
During the MPN Hub Steering Committee Meeting in May 2026, Andreas Reiter, University Hospital Mannheim, Mannheim, DE, delivered a presentation on targeting KIT in advanced systemic mastocytosis (AdvSM). In this presentation, Reiter discusses the rationale for KIT inhibition, and key efficacy and safety data for KIT-targeting therapies in AdvSM. The presentation highlights data from key trials, including the depth of molecular response with KIT-targeting therapies and its correlation with clinical benefit, and concludes with an overview of future directions in KIT inhibition for patients with AdvSM. This educational resource is independently supported by Blueprint Medicines. • Symposium | Advanced systemic mastocytosis: Clinical challenges and unmet needs

    17:23|
At the European Hematology Association (EHA) 2026 Congress, June 11–14, 2026, Stockholm, SE, the MPN Hub held a symposium, titled Advanced systemic mastocytosis: Redefining the disease, rethinking patient outcomes. During the symposium, Andreas Reiter, University Hospital Mannheim, Mannheim, DE, delivered a presentation on clinical challenges and unmet needs in advanced systemic mastocytosis (AdvSM).  In this presentation, Reiter discusses how genetic complexity underpins disease heterogeneity and clinical presentation in AdvSM, how variable disease presentation can lead to diagnostic delays, and that treatment outcomes highlight variability across AdvSM subtypes. He explores the limitations of conventional disease burden assessment, the variable symptom burden impacts on quality of life across systemic mastocytosis (SM) subtypes, and how disease heterogeneity can lead to complexity in AdvSM management. In addition, he discusses the variability in morphology and genetics in SM with an associated hematologic neoplasm, and illustrates potential challenges in clinical practice using a patient case study. This educational resource is independently supported by Blueprint medicines. • Symposium | Disease biology and molecular architecture in advanced systemic mastocytosis

    06:12|
At the European Hematology Association (EHA) 2026 Congress, June 11–14, 2026, Stockholm, SE, the MPN Hub held a symposium, titled Advanced systemic mastocytosis: Redefining the disease, rethinking patient outcomes. During the symposium, Deepti H Radia, Guy's and St Thomas' NHS Foundation Trust, London, UK, delivered a presentation on disease biology and molecular architecture in advanced systemic mastocytosis (AdvSM).  In this presentation, Radia discusses the classification and biology of AdvSM, covering mast cell disease biology, the clinical and biological spectrum of AdvSM, KIT as a central molecular driver of AdvSM, additional somatic mutations, clonal architecture and disease evolution, and the mechanisms of disease progression and organ damage. This educational resource is independently supported by Blueprint medicines. • ASH 2025 | Nuvisertib + momelotinib in patients with R/R MF: Phase I/II findings

    03:37|
During the 67th American Society of Hematology (ASH) Annual Meeting and Exposition, December 6–9, 2025, Orlando, US, the MPN Hub was pleased to speak with Claire Harrison, Guy’s and St Thomas’ NHS Foundation Trust, London, UK. We asked, What are the latest findings from the phase I/II study of nuvisertib in combination with momelotinib in patients with relapsed/refractory myelofibrosis (R/R MF)?In this interview, Harrison discusses key findings from Arm 3 of the trial (NCT04176198), which enrolled patients who had been previously treated with a Janus kinase inhibitor (JAKi) other than momelotinib. The study aimed to identify the recommended phase II dose (RP2D) of nuvisertib when administered in combination with momelotinib, and to assess the safety, clinical activity (spleen volume reduction [SVR] and total symptom score [TSS] improvement), and pharmacokinetics and pharmacodynamic markers (including cytokines and bone marrow fibrosis). This educational resource is independently supported by Sumitomo.
